No previous studies have investigated the use of graphite instead of activated charcoal in orchids. In this work, different concentrations of activated charcoal or graphite were added to KC medium to darken the culture medium and stimulate the in vitro propagation of Cattleya bicolor and of a double hybrid orchid (‘BLC Pastoral Innocence’). The seedlings were inoculated on growth regulator-free KC medium; the effects of activated charcoal (0.0, 1.5, 3.0, 4.5, 6.0, and 7.5 g L-1) and graphite (0.0, 1.5, 3.0, 4.5, 6.0, and 7.5 g L-1) concentrations were tested separately. Each flask contained fifteen seedlings, and the experiments had a randomised, two-way factorial design. The two tested variables were culture medium and illumination levels; eleven culture...

Development of a bioassay to quantify the ricin toxin content of castor bean (Ricinus communis L.) seeds - doi: 10.4025/actasciagron.v34i4.11284 Agronomy
Demant, Carlos Alberto Rauer; Universidade Estadual Paulista “Julio de Mesquita Filho”; Auld, Dick; Texas Tech University, Lubbock, Texas, United States; Demant, Aline Rodrigues de Mello; Texas Tech University, Lubbock, Texas, United States.
In this study, we developed a bioassay to quantify the ricin toxin content of castor bean seeds. Existing quantification methods do not always reflect actual toxicity of the seeds analyzed, which may present lower ricin content even though they are more toxic than seeds presenting a higher content of ricin. This is because these methods actually measure the addition of ricin RCA, which is a compound less toxic than pure ricin. We decided to use in this study, the nematode Caenorhabditis elegans, which has been widely used by the pharmaceutical industry. We tested two strains of C. elegans using different methods in 8 experiments. We examined 4 methods of extracting the ricin complex and 3 methods of exposing the nematodes. Among the nematode strains and...

The first step in plant transformation using somatic embryogenesis or organogenesis for plant regeneration in vitro has been the identification of genotypes with the best totipotency. Thus, the objective of this experiment was to evaluate the totipotency of ten cultivars and one soybean line from the Coodetec Gene Bank. The somatic embryos developed from immature cotyledons measuring from 4 to 6 mm in length, and the organogenesis was evaluated from mature soybean seeds. The cultivars CD 201, CD 216 and CD 220 were the most promising genotypes to produce somatic embryos, whereas the cultivar CD 217 had the highest quantity of differentiated explants, which represents the best regeneration efficiency for participating in organogenic systems. In the...

Influences of presowing treatments on the germination and emergence of fig seeds (Ficus carica L.) - doi: 10.4025/actasciagron.v34i3.13392 Agronomy
Çalişkan, Oguzhan; Faculty of Agriculture, University of Mustafa Kemal; Mavi, Kazim; Faculty of Agriculture, University of Mustafa Kemal; Polat, Aytekin; Faculty of Agriculture, University of Mustafa Kemal.
Seed germination of fig seeds is important for obtain seedlings with high variability in breeding studies, and for producing mosaic virus-free seedlings. This experiment was carried out to evaluated the effects of several treatments (priming with water for 24h, GA3 at 500 and 1000 ppm for 24h, 3% KNO3 for 24h, and stratification at 4°C for 7, 14 and 21 days) on seed germination and emergence of ‘Bursa Siyahı’ and ‘Sarılop’ fig cultivars. The application of GA3 increased the germination and emergence of these fig seeds. The highest percentages of germination and emergence were obtained with GA3 at 500 or 1000 ppm for both cultivars. The seed germination and emergence of the cultivar ‘Bursa Siyahı’ were higher than observed with the ‘Sarılop’. The...

In Iran, a large number of Rosa species have been exposed to extinction and therefore preservation techniques are necessary to safeguard their future. In the present investigation, the objectives were to optimize the micropropagation and medium-term conservation of one of these wild species, Rosa pulverulenta. At proliferation stage, the maximum number of new leaves (9.6) were produced on the medium containing 4 µM BAP + 0 µM GA3, whereas the maximum number of axillary shoots (4.1) were observed in the medium containing 4 µM BAP + 3 µM GA3. The results for rooting experiments suggested that the highest increase in stem height (49.5 mm) at the acclimatization stage was observed in plantlets treated with 1 µM...

Morphological and ultrastructural analysis of various types of banana callus, cv. Prata anã - doi: 10.4025/actasciagron.v34i4.14501 Agronomy

This work was carried out to characterise morphologically and ultrastructurally the banana callus, obtained from the scalp method. Genotypes of banana, cv. Prata anã, cultivated in vitro were used to induce meristems at the leaf base; subsequently, structures known as scalps were formed. For ultrastructural analysis, five samples of callus were collected, fixed in Karnovsky solution and analysed by scanning electron microscope (SEM) and transmission electron microscope (TEM). The formation of three types of callus was observed: Type 1 - transparent watery callus, Type 2 - yellow callus with small clusters, Type 3 - yellow callus with large clusters. SEM analysis showed that Type 1 callus cells were elongated and that Type 2 and Type 3 callus cells were...

Use of complex supplements and light-differential effects for micropropagation of Hadrolaelia purpurata (= Laelia purpurata) and Encyclia randii orchids - doi: 10.4025/actasciagron.v34i4.12333 Agronomy

In the current study, complex (coconut water and ripe-banana pulp) and specific (micronutrients, peptone and activated charcoal) supplements were added to Knudson-C medium (KC) medium to stimulate the in vitro propagation of Hadrolaelia purpurata (=Laelia purpurata) and Encyclia randii orchid species. The highest number of H. purpurata seedlings occurred in KC medium containing 90 g L-1 banana pulp maintained with 24-hour illumination. Positive effects of the banana pulp addition in KC medium were reported for the formation of seedlings and leaves from E. randii seeds. The seedling number from seeds of E. randii maintained with continuous illumination was also high in KC medium containing coconut water. Positive effects due to the addition of peptone were...

Fogaça, Fabiola Helena dos Santos; Embrapa Meio-Norte; Oliveira, Elenise Gonçalves de; Universidade Federal do Ceará; Carvalho, Samanta Eulles Quixaba; Embrapa Meio-Norte; Santos, José Francisco de Seixas; Embrapa Meio-Norte.
The fillet yield and quantity of waste generated after processing are important for planning and evaluating the growth performance of species and systems of aquaculture production in Brazil. The pirarucu (Arapaima gigas) is among the species with desirable characteristics for intensive fish with high market value. The objective was to evaluate the performance of pirarucu fillet in different weight classes: 7.0 to 9.0 kg (group I), 11.0 to 13.0 kg (group II) and 14.0 to 17.0 kg (group III). The fillet yield presented no significant difference between groups. The composition showed significant differences (p < 0.05) for moisture (77.96 to 75.04%) and ash (2.21 to 2.46%), while values of lipid and protein were similar between classes evaluated. In...
